放免法用于检测基因工程防龋疫苗抗体的研究

摘　　要：用国产12 5I同位素 ,以氯胺T氧化法 ,标记变链菌GTF抗原 ,其放射碘利用率 >40 % ,放化纯度 >95 % ,所得标记物置 - 2 0℃ 12 0d或 4℃ 6 0d仍可使用。采用放免法检测基因工程防龋疫苗免疫的家兔、鸡血清抗GTF抗体和卵黄抗GTF抗体 ,结果提示 ,该法是一种特异性强 ,灵敏度高 。A RIA was established by using 125 I isotype as indicator labelled to Streptococcus GTF antigen by chloramine toxidation,the ratio of 125 I utilization >40% and the purity of radioactivity >95%.The obtained label wre stored at -20℃ for 120 days or at 4℃ for 60 day,which are still applicable.The antibodies to GTF were detected both in the serum and eggs of hens by RIA method.The result showed good specificity sensitivity,resproducibility and stability of RIA method.
