柱串联法在凝胶过滤色谱中的应用一例

摘　　要：凝胶过滤色谱因其操作方法简单、重复性强等特点在大分子的分离纯化中被广泛使用 ,特别在基因工程蛋白质类药物的精细纯化中起着难以替代的作用。将Superdex 75凝胶柱与SephacrylS 10 0凝胶柱串联在一起进行凝胶过滤 ,分离一种用普通凝胶过滤色谱难以分离的样品 ,成功地将分辨率 (Rs)由 0 .71提高到 1.70 ,得到了基线分离。Gel filtration chromatography has occupied a key position in the purification of macromolecules for its simplicity and high reproducibility, especially in the polishing purification step of gene engineering protein drugs. In this article, a gel filtration column packed with Superdex 75 prep grade was connected in series with another filtration column packed with Sephacryl S 100 HR. A highly complex sample which was difficult for purification with normal gel filtration was applied on this two columns in series, and a rise of resolution from 0.71 to 1.70 occurred so that a baseline separation reached.
