摘 要:目的 :研究血管内皮生长因子 (VEGF)反义RNA对人脑垂体瘤细胞的抑制作用 ,探讨以VEGF反义RNA进行垂体瘤基因治疗的可行性。方法 :将VEGF反义cDNA、正义VEGFcDNA质粒转染垂体瘤细胞 ,通过Northern印迹杂交鉴定其表达 ,测定被转染细胞生长率 ,用原位杂交和免疫组化法检测VEGFmRNA及蛋白表达。结果 :经鉴定被转染的垂体瘤细胞有外源性VEGF反义基因的整合及表达 ,其VEGFmRNA及蛋白表达水平降低 ,但其生长速率无明显减慢。结论 :VEGF在垂体瘤血管形成及发生、发展中起重要作用 ,可能成为基因治疗垂体瘤的优选靶的之一。Aim:To study the inhibitory effect of VEGF antisense RNA on the growth of human pituitary adenoma cells in vitro, in order to further investigate the feasibility of VEGF antisense RNA gene therapy of pituitary adenomas. Methods:Carrying with VEGF antisense cDNA, VEGF sense cDNA plasmid was transfected into pituitary adenoma cells, and confirmed its expression by Northern blot.The survival rate of transfected cells was detected by MTT assay.The expression of VEGF mRNA and protein was detected by Northern blot analysis, in situ hybridization and immunohistochemistry. Results:The expression of exogenous antisense VEGF mRNA was confirmed in transfected cells, and the endogenous VEGF mRNA and VEGF protein level was dramatically decreased. Conclusion: VEGF may play an important role in angiogenesis and development of pituitary adenoma, and suggests that VEGF gene is a target for gene therapy of pituitary adenoma.
