摘 要:将天蚕抗菌肽B基因应用基因枪法导入水稻TN1发芽种胚 ,获得转基因植株 2株 .转基因水稻T1~T55个世代不同株系 ,对bar基因、抗菌肽基因的遗传和表达进行初步研究 .结果表明 :转基因后代株系barR∶barS 大部分符合 3∶1分离比率 .T1、T4 转基因株系PCR、Southern印迹分析表明 :T18株有抗菌肽B基因 ;T4 仍有 4个株系有抗菌肽基因 ,其他的株系基因丢失 .T3 1个株系Northern印迹证明抗菌肽基因在RNA水平有表达 ;抗菌肽B转基因水稻对白叶枯病的抗病性有提高 ,抗性能传递到高世代 ,在T3Two transgenic plants transformed with cecropin B gene using biolistic method were generated from germinative embryo of rice variety TN 1. The inheritance and expression of bar and cecropin B gene in different transgenic plantlines of TN 1 from 5 generation of T 1 to T 5 were primary studied. Results showed that the segregation ratio of barR:barS was 3∶1 . The results of PCR and Southern blot analysis indicated that eight of T 1 and four of T 4 transgenic lines had cecropin B gene, the other plantlines of T 4 showed lost of cecropin gene. One of T 3 transgenic line expressed at mRNA level by Northern blot. The resistance to bacterial blight delivered from cecropin B transgene could be segregated and transmitted to high generation, resistance improved plantlines were obtained in T 3 generation.
