摘 要:目的 观察茶叶的主要成分茶多酚 (Teapolyphenol,TP)对 HL- 60细胞 DNA中 8-羟基鸟嘌呤 (8- oh- G)变化的影响。方法 利用气相色谱仪 (GC/ FID)、气质联用仪 (CGC/ MS- SIM)测定 DNA中 8-羟基鸟嘌呤含量 ,同时观察细胞丙二醛 (MDA)含量、还原型谷胱甘肽 /氧化型谷胱甘肽 (GSH/ GSSG)比值变化。结果 TP作为抗氧化剂在一定的浓度范围可以减少 DNA的氧化损伤产生 8-羟基鸟嘌呤的含量 ,但当 TP高达到一定浓度时反而增高 8-羟基鸟嘌呤的含量。结论 TP对细胞 DNA氧化损伤有抑制和促进两方面作用 ,这与 TP的浓度有关 ,即一定浓度时 TP具抗氧化作用 。? Objective To study the effect of the teapolyphenol on amount of 8 hydroxyguanine in Hl 60 cellular DNA that were damaged by H 2O 2.Methods Amount of 8 hydroxyguanine in DNA was detected by GC/FID and CGC/MS SIM.Simultaneously the ratio of GSH/GSSG and the level of MDA were measured.Results The number of 8 hydroxyguanine has reduced in DNA,the amount of MDA has decreased,the ratio of GSH/GSSG has increased in cells when the concentrations of teapolyhphenol were 10 mg·L -1 ,however the number of 8 hydroxyguanine in DNA, the level of MDA in cells has increased and the ratio of GSH/GSSG in cells has lowered when the concentrations of teapolyphenol attended 40 mg·L -1 .Conclusions Teapolyphenol can affect on the amount of 8 hydroxyguanine in HL 60 cells that were damaged by H 2O 2.It depends on the concentrations of teapolyphenol,the lower concentrations of teapolyphenol inhibits the oxidization of H 2O 2 while the higher concentrations of teapolyhenol can enhances the result of oxidization. 〔
