摘 要:目的 观察雌激素和胆酸负荷对未孕大鼠肝胆功能及有关激素水平的影响。方法 将 37只近交系Wistar大鼠随机分为三组 ,试验Ⅰ组肌肉注射苯甲酸雌二醇 (溶于丙三醇 ) ,试验Ⅱ组静脉注入游离胆酸 ,对照组肌肉注射丙三醇。所有动物于胆汁收集前后采血 ,分离血浆。测定血浆与胆汁中总胆酸 (TBA)、甘氨胆酸 (CG)、总胆红素 (TBil)、丙氨酸转氨酶 (ALT)、天冬酸转氨酶(AST)、碱性磷酸酶 (ALP)、γ 谷氨酰转肽酶 (GGT)、总胆固醇 (TC)、雌二醇 (E2 )等肝胆功能指标及激素水平 ,并作统计分析。结果 雌激素注射后大鼠胆汁分泌量显著减少和胆流速度显著减慢。雌激素试验Ⅰ组与对照组相比 ,胆汁收集前血浆中GGT、E2 显著升高 ,TC显著下降。胆汁收集后血浆中ALP、GGT、E2 显著升高 ,TC显著下降。胆汁中TBil、ALP、GGT、TC较对照组显著升高 ,TBA显著下降。试验Ⅱ组与对照组相比 ,胆汁收集前血浆中GC、TC、ALP显著升高 ,T1Bil、E2 、ALT、AST显著下降。胆汁收集后血浆中GC、ALP显著升高 ,ALT、TBil显著下降。胆汁中GC、ALP、GGT显著升高 ,TBil显著下降。胆酸注入组较E2 注射组胆汁中TBA、GC显著升高 ,TBil、ALP、TC显著下降。结论 雌激素可降低胆汁分泌功能 ,引起肝内淤胆 ,但不直接损伤肝细胞。雌激素可导致胆固醇?Objective To observe how cholic acid and estrogen loading affects liver and gallbladder function and some hormones in dry rats. Methods Inbred Wistar rats,37,were divided randomly into three groups.Group I were injected with benzogynestryl dissolved in propyl alcohol by i.m., 5 mg/kg body weight,for five day.Group Ⅱ received intravenous infusion of cholate.The control group were injected only with propanetrid.Blood samples were collected before and after the bile collection,and plasma was separated.TBA,CG,TBil,ALT,AST,ALP,GGT,TC and E 2 were determined.Results After the rats were injected benzogynestryl,flow of bile secreation decreased significantly.Compared with the control group,plasma GGT and E 2 of Group I increased and TC decreased significantly before bile was collected,and ALP,GGT and E 2 increased and TC decreased significantly after bile was collected.Billiary TBiL,ALP,GGT and TC increased and TBA decreased significantly.Plasma CG,TC,ALP of Group Ⅱ increased and TBil,E 2,ALT and AST decreased significantly before bile was collected, and CG and ALP increased,and ALT and TBiL decreased significantly after bile was collected,biliary CG,ALP and GGT increased and TBA decreased significantly.Compared with Group I,biliary TBA and CG of Group Ⅱ increased and TBil,ALP,Tc decreased significantly.Conclusion Estrogen can decrease bile secretion,resulting in intrahepatic cholestasis,and does not cause direct damage to the liver cells.Estrogen can decrease TC synthesis,but does not alter concentration of plasma cholic acid.Cholate can increase concentration of plasma cholic acid,leading to intrahepatic cholestasis,decrease plasma estrogen,and does not cause direct damage to liver cells.
