摘  要:目的 :探讨碱性成纤维细胞生长因子 (bFGF)对人牙周膜细胞的增殖与碱性磷酸酶 (ALP)活性表达的影响 ,为bFGF应用于牙周组织的再生与修复提供实验依据。方法 :采用MTT和ALP活性检测法 ,观察不同浓度 (0 1~ 10ng/ml)的bFGF第 2 4、48和 72小时对 5 %和 10 %胎牛血清 (FBS)体外培养的第 3代人牙周膜细胞 (PDLCs)的作用。结果 :与对照组比较 ,① 10 %FBS ,5ng/ml和 10ng/ml的bFGF在第 2 4、48、72小时 ,可显著促进PDLCs的增殖 (P <(0 0 1~ 0 0 5 ) ,5 %FBS ,10ng/ml的bFGF在 2 4、48小时 ,5ng/ml的bFGF在 2 4小时对PDLCs有显著促增殖作用 (P <0 0 1)。② 10 %FBS ,1ng/ml的bFGF在 48、72小时 ,5ng/ml的bFGF在 2 4、48、72小时 ,10ng/mlbFGF在 48小时对PDLCs有显著增强ALP活性的作用P <0 0 1— 0 0 5 ) ;5 %FBS ,10ng/ml的bFGF在 2 4、72小时对PDLCs有显著增强ALP活性的作用 (P <0 0 5 )。结论 :以上结果提示 :在一定的作用时间内和一定条件下 ,5ng/ml和Objective:[WT5”BZ]To evaluate the biological effects of basic fibroblast growth factor(bFGF)on the proliferation and alkaline phosphatase(ALP)activity in 5% and 10% FBS cultured human periodontal ligament cells(PDLCs).[WT5”HZ]Methods:[WT5”BZ]Thiazoly blue(MTT)colorimetric assay and ALP activity analysis were used at different time points(the 24th,48th and 72th hours)to determine the proliferation and ALP activity of PDLCs after different doses of bFGF were added into the culture medium.[WT5”HZ]Results:[WT5”BZ]When compared with control group,①10%FBS,5ng/ml and 10ng/ml bFGF at the 24 th ,48 th and 72th hours,significantly promoted the proliferation of PDLCs(P<0 01~0 05);5%FBS 10ng/ml bFGF at 24 th and 48 th hours,5ng/ml bFGF at 24 th hours,significantly stimulated the proliferation of PDLCs (P<0 01);②10% FBS 1ng/ml bFGF at 48th and 72th hours,5ng/ml bFGF at 24 th ,48 th and 72th hours,10ng/ml bFGF at 48 th hours,significantly increased the ALP activity of PDLCs(P<0 01~0 05);5%FBS,10ng/ml bFGF at 24 th and 72th hours significantly increased the ALP activity of PDLCs(P<0 05).[WT5”HZ]Conclusion:[WT5”BZ]These results suggested that 5ng/ml and 10ng/ml bFGF may simultaneously promote the proliferation and differentiation of human PDLCs during a period of time under specified condition.
