摘 要:应用荧光原位杂交 (FISH)技术评价了昆明山海棠水抽提物 (THH)在雄性小鼠生殖细胞形成过程中的非整倍体诱发效应。以THH(1 2 0mg/kg,2 4 0mg/kg ,4 80mg/kg)腹腔注射昆明种雄性小白鼠 ,根据小鼠精子发育周期 ,于处理后 2 2天后取小鼠附睾精子涂片 ,以生物素标记的 8号染色体着丝粒重复顺序探针进行FISH ,荧光显微镜下分析受试物处理动物后精子特异染色体分离情况。结果发现 ,THH在中间剂量组 (2 4 0mg/kg)和高剂量组 (4 80mg/kg)诱发精子非整倍体频率均显著高于溶剂对照 (P<0 .0 0 1 ) ,低剂量组 (1 2 0mg/kg)的异常精子频率与溶剂对照无显著差异 (P >0 .0 5)。研究证明 。Fluorescence in situ hybridization (FISH) was to employed to evaluate the inductive effect of aneuploid in male mice sperm after treated by Tripterygium hypoglaucum(Levl.) Hutch (THH). The animals were given THH (120, 240, 480mg/kg) and colchicine(COL) by abdominal cavity injection, according to developmental cycle of mice sperm, killed animals and made smear after 22 days treatment. FISH was applied to detection frequencies of aneuploid with biotin labelling DNA probes specific for chromosome 8. The aneuploid frequencies were significantly higher than the solvent control (p<0.001)in 240mg/kg group and 480mg/kg group. The aneuploid frequencies were not significantly different between the sperm in 120mg/kg group and solvent control (p>0.05). It is conclude that THH is a potential germinal aneugen in mouse sperm.
