摘  要:采用 RT- PCR方法克隆了黄瓜花叶病毒西番莲致死分离物 ( CMV- PE)全长 RNA3.经核苷酸序列测定 ,明确 PE分离物 RNA3全长 2 2 16nt,含有 2个开放阅读框 ( ORF) ,其中 5'端的 ORF( 12 1- 963nt)编码2 79aa的 3a蛋白 ,3'端 ORF( 12 60 - 1916nt)编码 2 18aa的 CP蛋白 .5'端非编码区 ( NR)长 12 0 nt,基因间隔区 ( IR)长 2 96nt,3'端 NR区含 30 1个碱基 .PE分离物编码的 3a蛋白中最明显的特征是在 136- 141位有一个独特的 VWCL SS区域 .将 CMV- PE的 RNA3的核苷酸序列及其编码蛋白的氨基酸序列与同属 CMV亚组 I的其它分离物进行比较 ,发现症状相似的 CMV分离物的非编码区具有很高的序列同源性 。The full length cDNA of RNA3 of CMV PE, an isolate of Cucumber mosaic virus from passionflower, was cloned and sequenced by reverse transcription PCR with two primers. The nucleotide sequence of RNA3 of CMV PE was composed of 2 216 nucleotides (nt) with two open reading frames, in which the one near 5'terminal (121 963 nt) encoded a 3a protein with 279 amino acids (aa), the other one near 3' terminal (1 260 1 916 nt) encoded a coat protein with 218 aa. The non coding regions (NR) located at 5'terminal and 3'terminal were 120 nt and 301 nt, respectively, and the intergenic region (IR) was 296 nt long. There was an unique protein motif with VWCLSS sequence located at the position of 136 141 of the 3a protein. When the RNA3 of CMV PE was compared with other isolates which also belong to subgroup I of CMV, those isolates which appeared similar symptom in hosts showed higher identity of nucleotide sequences of NR, therefore, the NR might be related to the symptom.
