摘　　要：激素敏感脂肪酶（ＨＳＬ）是动物体脂肪分解的关键酶，第８外显子编码着影响激素敏感脂肪酶活性的调节功能区。本研究合成引物ＰＣＲ扩增第８外显子部分片段，ＰＣＲ产物经ＳＳＣＰ分析，结果有多态性。该ＤＮＡ片段经过克隆测序分析，发现位于基因第２３０１处发生碱基序列的改变 （Ｇ→Ｔ），使编码的氨基酸由谷氨酸变为天冬氨酸（Ｇｌｕ→Ａｓｐ）。本研究还分析了多个品种在该座位的等位基因频率。Hormone-sensitive lipase (HSL) plays a key function of adipose metabolism in animal, and the exon Ⅷof hsl gene encodes a regulatory bioactive domain of this protein. The primers targeting to the exon Ⅷ were synthesized for PCR-SSCP analysis. The amplified DNA fragments were cloned and sequenced, and one substitution mutation (G →T) at the position 230l of this gene was identified,which lead to one amino acid replacement(Glu →Asp).The gene frequency at this locus in 4 purebreds of pig was also analyzed.
