摘  要:2114是一个携带有来自小伞山羊革的恢复基因的T型恢复系,为研究该恢复系恢复基因的遗传,我们将恢复系2114与 3个不育系 ND44A、D8442A、D北 15A杂交,通过调查三个组合的 F2代群体育性分离对恢复基因进行了遗传分析。分析表明除了一对主效基因控制外,2114育性同时又受微效基因的影响,而且2114中携带有恢复基因的易位染色体在不同遗传背景中雄配子的传递串不同。以ND44A/2114的F2代分离群体为作图群体,利用分离群体分组分析法(Bulked SegregantAnalysis,BSA),用460个随机引物对21 14中的T型细胞质雄性不育恢复基因Rf6进行RAPD分析,筛选到10个可在亲本间扩增出多态性的引物,其中OPI18经多次重复能在亲本间及不育和可育池间扩增出稳定的多态性片段OPI181260。进一步对F2群体的 147个单株分析表明:OPIl81260与恢复基因 Rf6的遗传距离为 3.4 cM。It's very important to develop new fertility restorer line in hybrid wheat breeding. 2ll4 is a restorer line which contained one restorer gene, Rf6, derived from Aegiiops umbellulata Zhuk.. Aiming to further understand the inheritance of fertility restorer gene Rf6, three sterile lines ND44A, D8442A, D Bei l5A were used as female to cross with 2ll4. The analysis of the 3 F1.s indicated that the restoring ability of 2l l4 is over 70% in green house condition and 86.4% in field condition in Beijing. The segregation ratio of fertile to sterile in F2populations is diverged to 3; l due to the abnormal transmission frequency of the translocation chromosome and other modified factors. Bulked Segregan Analysis was employed to identify Random Amplified Polymorphic DNAs (RAPD) markrs linked to the restorer gene (Rf6) in the F2population of ND44A/2114. Among the total 460 arbitrary l0-mer oligonucleotide primers screened, 10 produced polymorphisms between the parents. One of them, OPI18, was identified to be associated with Rf6. The polymorphic band was designed OPI181260. Further RAPD analysis was conducted in l47 F2 plants of ND44A/2114 by primer OPI18. It was found that the distance between Rf6 and OPIl81260. is 3.4 cM.
