摘  要:目的 探讨脑梗死和偏头痛患者血小板内钙浓度及其凝血酶刺激后钙浓度的变化。方法 应用Fura 2负载血小板双波长扫描 ,连续测定了 2 0例脑梗死患者与 2 0例偏头痛患者血小板静息态胞浆游离钙浓度 ([Ca2 + i])及观察凝血酶诱导后血小板 [Ca2 + i]动态变化 ,并记录了凝血酶刺激后血小板 [Ca2 + i]上升时间 (t)。结果 脑梗死患者和偏头痛患者血小板静息态、凝血酶诱导后 [Ca2 + i]及凝血酶诱导后较静息态的[Ca2 + i]净增量△ [Ca2 + i]均显著高于各自的对照组。脑梗死患者和偏头痛患者比较 ,上述指标差异不显著 ;脑梗死组凝血酶刺激后血小板 [Ca2 + i]上升时间较对照组明显延长 ,而偏头痛组较对照组明显缩短 ,脑梗死组较偏头痛组延长。结论 血小板 [Ca2 + i]异常增高可能是该两种疾病血小板异常的共同机制之一 ,血小板受刺激后 [Ca2 +Objective To explore the difference of concentrations of platelet cytosolic ionized calcium([Ca 2+ i]) in the basal state and following stimulation with thrombin. Methods A calcium sensitive fluorophor Fura s with dual excitation wave length was used to measured the concentrations of platelet cytosolic ionized calcium([Ca 2+ i]) in the basal state and following stimulation with thrombin in 20 cerebral thrombosis and 20 migraine patients. Results Compared to the contrals,basal and stimulus induced[Ca 2+ i] were high in platelets of both patiel groups. There were no apparent significant differences in platelet[Ca 2+ i] levels between both patient's groups.However, the speed to reach the maximum[Ca 2+ i] level after stimulation by thrombin was rather slower in cerebral thrombosis patients,and more rapid in migraine patients than in the non diseased contronls. Conclusion It is suggested that the increase of cytosolic[Ca 2+ i] in platelets following thrombin stimulation should be one of the common mechanism of migraine and cerebral thrombosis the difference of time to attain the maxium level of platelet[Ca 2+ i] levels following thrombin activation between them may reflect the difference of mechanism of two diseases in natural.
