树突状细胞体外产生特异性抗白血病免疫应答的研究

摘  要:目的 探讨白血病患者外周血树突状细胞 (DCs)在体外诱导抗白血病免疫反应。方法 用急性早幼粒细胞白血病 (M3)细胞株 (HL - 6 0 )的肿瘤相关抗原 (TAA)激活DCs,以粒 /巨噬细胞集落刺激因子 (GM -CSF)及白介素 - 4(IL - 4)联合刺激M3 患者外周血DCs,在体外诱导自体混合T淋巴细胞增殖。分化为细胞毒性T细胞 (CTL)。结果 该CTL及其上清液对HL - 6 0白血病细胞有高效、特异的杀伤作用 ,而对慢性粒细胞白血病细胞株 (K 5 6 2 )及肝癌细胞HepG2 仅有微弱的杀伤作用。结论 DCs能有效提呈肿瘤抗原并诱导出显著的杀伤白血病细胞的免疫反应 。Objective To discuss antileukemic immune response induced by dentritic cells (DCs) in peripheral blood of leukemic patients.Methods DCs were activated by tumor associated antigen(TAA) from acute granulocytic leukemic(M3) cell line (HL-60).DCs in peripheral blood of leukemic patients were stimulated by granulocyfe-macrophage cell stimulating factor (GM-CSF) and interleukin-4(IL-4).T-cells were induced in vitro to proliferate and differentiate into cytotoxic T lymphocytes (CTLs).Results CTLS could destroy HL-60 leukemic cells effectively and specifically,whereas they only had weak killing effects on chronic granulocytic leukemic cell line(K 526) and hepatic carcinoma cells(HepG2).Conclusions DCs can effectively present tumor antigen and induce immune reaction to kill leukemic cells,which may provide a new approach to tumor specific immunotherap.
