摘  要:目的  研究足叶乙甙 (VP16 )对K56 2细胞分化的诱导作用。方法  K56 2细胞在含0 .4 μg mlVP16的培养液中培养 72h ,观察K56 2细胞NBT还原能力和吞墨功能 ,以及细胞化学和超微结构的变化。结果 0 .4 μg mlVP16可使K56 2细胞NBT还原能力和吞墨功能增强 ,细胞内过氧化物酶、糖原和α 萘酚酯酶含量增加 ,电镜下可见细胞体积变小 ,微绒毛和线粒体减少 ,核异染色质增加。结论 VP16诱导K56 2细胞向粒单细胞系分化。Objective To explore the effect of VP16 on the induction of differentiation of K562 cells.Methods K562 cells were left to grow for 72 hours in the culture solution which contained 0.4ug/ml VP16.Observations were made to know about the NBT reducing power,the functions of absorbing prepared Chinses ink of K562 cells,the cytochemical changes and the ultrastructural changes.Results 0.4ug/ml VP16 can effectively enhance the NBT reducing power and the function of absorbing prepared Chinese ink of K562 cells,increase the content of intracelluar peroxidase,glycogen and α naphtolate esterase.Through electron microscope,the shrinking of cell volume,the decrease of microvillui and mitochondria,and the increase of nuclear heterochromatin could be observed.Conclusion K562 cells can be induced and differentiated into granular unicellular line.
