摘  要:以环氧氯丙烷活化的磁性壳聚糖微球作为载体,对脲酶进行固定化研究。结果表明, 在25℃时, 活化磁性壳聚糖微球对脲酶的固定化在2h时就达到了最大值,固定化酶和自由酶 的最适温度都在65℃左右,自由酶和固定化酶的米氏常数Km值分别为0.042mol/L和0.008mol /L,固定化酶的Km降低了5倍。The magnetic chitosan microsphere were activated by epichlorohydrin in the presence of sodium hydroxide, which were used as carrier for the immobiliza tion of urease. It was found that the content of epoxy in the activated magnetic chitosan microsphere was 0.65mmol/g. The immobilizated urease will be obtained at 2.0h. The optimum temperature of the immobilizated urease was close to that o f the native urease. The half times for the decay of activity were about 2.0h fo r the immobilization urease and native urease. The optimum pH of the immobilized urease decreased by 0.4. The Km value of immobilized urease and native urease a re 0.008mol/l and 0.042mol/l, respectively.
