摘  要:目的 研究维甲酸对大鼠胃粘膜癌变DNA含量、c myc基因mRNA表达及细胞凋亡的影响。 方法 采用MNNG、酒精及 10 %NaCl混合液建立大鼠胃癌模型 ,设立正常空白组、模型组及维甲酸治疗组 ,分别采用Feulgen染色、原位杂交及TUNEL法检测大鼠胃粘膜DNA含量、c myc基因表达及细胞凋亡。 结果 治疗组癌变发生率低于模型组。c myc基因mRNA表达与细胞凋亡受抑制和胃粘膜癌变的发生 ,呈递增趋势 ,并且两者具有一定的相关性。结论 维甲酸抗癌 ,下调c myc基因表达 ,促进细胞凋亡是其重要机制之一。Objective To study the effects of transretinoic acid on DNA, cMYC mRNA exepression and apoptosis of gastric cancer in rats. Methods we studied DNA , expression of cMYC gene mRNA and apoptosis using feulgen, in situ hybridization, TUNEL in rats in three groups: 1.Normal control,2.Model control,3.Treatment groups with transretinoic acid in gastric cancer model induced by MNNG,alcohol and 10% sodium chloride.Results The incidence of gatric cancer in trenment groups was lower than that of model ones. With the development of gastric cancer, the expression of cMYC gene and supression of apoptosis were increased. A corrolation was present between both.Conclusion Transretinoic acid can both downregulated the expression of cMYC gene and promote cell apoptosis.
