摘 要:采用原位杂交技术及放免分析法观察了 35只SD大鼠应激时下丘脑、垂体前叶、肾上腺组织中血管紧张素原(ANG)mRNA的表达及血管紧张素II(AngII)含量变化。结果表明 :下丘脑、垂体前叶、肾上腺髓质ANGmRNA表达均显著增强 (n =5 ,P <0 0 0 1,P <0 0 5 ) ,以下丘脑最为明显。下丘脑、垂体前叶ANGmRNA表达增高率超过了AngⅡ含量的增高率 (P <0 0 5 ) ,而肾上腺皮质ANGmRNA表达仅在急性应激时有明显改变 (P <0 0 5 )。结果提示 :应激时局部组织肾素 血管紧张素系 (RAS)被显著激活 ,以往测得的局部组织AngⅡ的增高主要是局部RAS激活的结果 。The expression of angiotensinogen (ANG) mRNA and the changes of AngII contents in hypothalamus, anterior pituitary, adrenal medulla and adrenal cortex of rats (n=35) during acute and chronic stress were investigated by in situ hybridization technique and RIA The results showed that the ANGmRNA expression was significantly increased in hypothalamus (n=5, P <0 001), anterior pituitary (n=5, P <0 05), and adrenal medulla (n=5, P <0 05), especially in hypothalamus The increased ratios of ANGmRNA surpassed the ones of AngII contents in hypothalamus and pituitary However, in adrenal cortex, ANGmRNA expression was only increased during acute stress ( P <0 05) In summary, our results suggested that the local tissue renin angiotensin system (RAS) was obviously activated during stress, and the increase of AngII contents in the local tissues during stress, which we previously observed, originated mainly from the activation of local RAS The present study strongly supports the view that local tissue RAS contributes to stress reaction
