摘　　要：[目的]研究CYP2E1RsaⅠ、GSTT1、GSTM1基因多态性和烟酒茶嗜好及其相互作用与食管癌易感性的关系。[方法]在食管癌高发区淮安市进行了一个病例对照研究(食管癌144例 ,人群对照233例) ,调查研究对象的烟酒茶嗜好习惯 ,以PCR扩增、RsaⅠ内切酶消化 ,分析CYP2E1的基因型 ,以多重PCR方法分析GSTT1、GSTM1基因型。[结果](1)对照组GSTM1基因型频度及饮茶状况分布与食管癌组之间存在显著差异 ,携带GSTM1正常基因型或有饮茶习惯者发生食管癌的危险性显著降低。(2)在不同烟酒茶嗜好者中 ,CYP2E1、GSTT1和GSTM1基因型对食管癌发生的影响有所不同。[结论](1)携带GSTM1正常基因型或饮茶能降低食管癌发生的危险性。(2)To evaluate the relationship among polymorphisms with CYP2E1 RsaⅠ, GSTT1 and GSTM1,habit of smoking,alcohol and tea,and their interaction on the risk of esophageal cancer. We conducted a case control study with 144 cases of esophageal cancer and 233 population based controls in Huaian city of Jiangsu province,China.The epidemiological data were collected,and DNA of peripheral blood leukocytes was obtained from all of the subjects.CYP2E1 genotypes were assayed by PCR amplification followed by digestion with RsaⅠ.Genotypes of the GSTT1 and GSTM1 were assayed by multiplex PCR method.(1) A significant difference was observed in the frequency of GSTM1 genotypes and in the habit of drinking tea between controls and esophageal cancer cases.Drinking tea and GSTM1 normal genotype significantly decreased the OR for esophageal cancer.(2) The roles of CYP2E1,GSTT1 and GSTM1 genes on the development of esophageal cancer were different based on the consumption of tobacco,alcohol and tea. [Conclusions]GSTM1 normal genotype or drinking tea may have a protecting effect against esophageal cancer.Interaction of CYP2E1,GSTT1 and GSTM1 genes polymorphism and the environmental factors exists in the oncogenesis of esophageal cancer.
