摘 要:目的 研究不同剂量苦参碱 (MT)对二乙基亚硝胺 (DEN)诱发大鼠肝癌的预防阻断作用。方法 采用0 0 1%的DEN诱发大鼠肝癌 90d ,同时分别用MT注射液30、15、3mg/kg腹腔注射和 3mg/kg灌胃。停止诱癌及给药处理 30d后 ,处死大鼠 ,观察大鼠肝脏的病理改变、肝表面癌结节数、肝 /体重比和血清中丙氨酸氨基转移酶 (ALT)、γ 谷氨酰转肽酶 (γ GT)、碱性磷酸酶 (ALP)的变化。结果 3mg/kgMT腹腔注射组和灌胃组的体重明显高于模型组 ,肝表面癌结节数、肝 /体重比和血清ALT、γ GT明显低于模型组和它MT腹腔注射组 (P <0 0 5 ) ;但腹腔注射的三组中ALP较模型组有不同程度升高 ,灌胃组则有所下降。结论 MT用于预防阻断DEN诱发大鼠肝癌时 ,不论剂量大小或给药途径如何 ,均不能完全阻断DEN诱发大鼠肝癌的发生。但小剂量 (3mg/kg)的MT长期口服应用 ,能保护肝细胞免受损伤 。Objective To study the preventive effect of different doses of matrine (MT) on rat hepatoma induced by diethylnitrosamine (DEN).Metho ds Rats with hepatoma induced by 0.01% DEN for 90 days, were administer ed with MT 30,15, 3 mg/kg ip and 3 mg/kg po at the same time. When 120 days have passed, the rats were killed and changes of hepatoma nodules, body weight, live r weight, ratio of liver/body weight were recorded and the alanine aminotranster ase (ALT), γ-glutamyl transferase (γ-GT) and alkaline phosphatase (ALP) in s erum and the pathological changes of the livers were observed. Results In the two groups of 3 mg/kg MT ip and po, the body weights were larger than that in the model group; the hepatoma nodules, the ratios of liver/body wei ght and serum ALT, γ-GT were lower than those in the model group and groups of 30, 15 mg/kg MT ip ( P <0 05). But the serum ALP of MT ip groups were highe r than that of the model group ( P <0 05),it was lower in the MT po group. Conclusion 3 mg/kg MT ip or po can protect liver cells form dam age induced by DEN and the development of hepatoma may be delayed. No matter wha t different doses and ways of MT are used. MT can not inhibit the production of hepatoma completely.
