摘 要:目的 研究p16基因启动子CpG区甲基化的改变与大肠癌发生及其临床病理特征的关系。 方法 采用甲基化特异PCR技术 (MSP法 )研究大肠癌组织 p16基因甲基化 ,并分析其与临床病理特征的关系。 结果 3 0例大肠癌组织中有 12例 (40 .0 % )呈 p16CpG区甲基化阳性。DukesC、D期组织 p16CpG区甲基化发生率明显高于DukesA、B期。有转移的大肠癌组织p16甲基化发生率 (84.6% )明显高于无转移者 (5 .9% )。结论 p16启动子区甲基化导致 p16抑癌基因失活 ,参与大肠癌的发生和发展。大肠癌组织 p16CpG区甲基化与Dukes分期、肿瘤转移有关。p16甲基化可作为评估大肠癌患者预后的Objective To explore whether methylation of the CpG island in the promotor of the p16 tumor suppressor gene is associated with development and clinicopathological characteristics of the colorectal cancer.Methods Methylation-specific PCR(MSP) was used to detect p16 methylation of the colorectal cancer specimen.Results In 30 sporadic colorectal cancer,40.0% had detectable p16 methylation.The patients with Dukes C,D were more likely to contain methylated p16 compared with those with Dukes A,B.The patients with tumor metastasis were more likely to contain methylated p16(84.6%) compared with those without tumor metastasis(5.9%).Conclusion p16 methylation plays a role in the carcinogenesis of colorectal cancer.Dukes stage and tumor metastasis is associated with p16 methylation status.p16 methylation might be considered as a prognostic indicator.
