摘 要:目的 :探讨 p16基因的异常表达与原发性肝癌发生、发展的关系。方法 :应用PCR、SSCP分析及DNA序列测定技术研究了 30例原发性肝癌组织中的 p16基因外显子 2 (E2 )的缺损情况。结果 :( 1)p16基因缺损的检出率与肝癌的肿瘤组织类型无关 ,在肝细胞癌与胆管细胞癌中p16基因缺损的检出率分别为 33 .33 %和 2 0 .0 0 % ,两组间无显著差异 (P >0 .0 5 ) ;( 2 ) p16基因缺损率的高低与组织学分级密切相关 (P <0 .0 5 )。结论 :结果提示p16基因的缺损与原发性肝癌的组织学分级相关 ,它可能参与原发性肝癌的发生。Purpose: To explore the relationship between the vacant status of the p16 gene and development of primary hepatocarcinoma. Methods: The PCR amplification of DNA, analysis of PCR products, the SSCP analysis and DNA sequencing were used to detect the loss of p16 INK4 gene exon 2 in 30 primary hepatocarcinoma tissues. Results: The positive rates of p16 gene deletion in hepatocarcinoma and hepatocholangiocarcinoma were 33.33% and 20.0%, and no significant difference was found among them(P> 0.05 ). The positive rates of p16 gene deletion was closely correlated with histological grade (P<0.05). Conclusion: Alteration of p16 gene is closely related to the histological grade. It may play an important role in the oncogenesis and development of the primary hepatocarcinoma.
