摘　　要：采用聚合酶链反应—变性凝胶高压电泳结合银染技术及基因序列方法 ,确认载脂蛋白B基因四核苷酸串联重复 (TTTA)n基因型 ,研究中国汉族人载脂蛋白B基因四核苷酸串联重复序列 (TTTA)n遗传多态性特征。结果 :汉族人群中 ,共检出载脂蛋白B四核苷酸串联重复序列 (TTTA)n有 3种等位基因和 6种基因型。基因型为 12 8/12 8,12 8/13 8,12 8/14 8,13 8/13 8,13 8/14 8,14 8/14 8,其频率分别为 0 0 0 5 ,0 0 89,0 0 10 ,0 816 ,0 0 75 ,0 0 0 5 ;等位基因为 12 8,13 8和 14 8,其频率分别是 0 0 5 5 ,0 898和 0 0 47,与美国高加索人相应的频率比较 ,两者的差异具有显著性 (P <0 0 1) ;结果说明 :载脂蛋白B基因四核苷酸串联重复 (TTTA)To explore the genetic polymorphism of apolipoprotein B tetranucleotide tandem repeat(TTTA)n in Chinese Han. The genotypes of the locus were discerned by polymerase chain reaction high voltage denaturing gel electrophoresis, silver staining and gene sequence analysis. Result:there were three alleles and six genotypes at the locus. The genotypes were 12.8/12.8,12.8/13.8,12.8/14.8,13.8/13.8,13.8/14.8,14.8/14.8. Their frequencies were 0.005,0.089,0,010,0.816,0.075,0.005, respectively. The alleles and their frequencies were 12.8(0.055),13.8(0.898)and 14.8(0.047), respectively. Comparing with that of Americal Caucasian, the alleles frequency distributions were significantly different( P <0.01). The results suggested that tetranucleotide tandem repeat(TTTA)n of apolipoprotein B gene was genetic polymorphism and heterogenous among population.
