表面铺展技术在联会复合体电镜研究中的应用

摘  要:本文较详细地介绍了本室改进的表面铺展 -硝酸银染色制备联会复合体 (SC)电镜样品技术。取小鼠睾丸组织制成细胞悬液 ,取少量悬液滴在 0 4 %KCl液珠表面上以通过表面张力使SC铺展开。用涂有Formvar膜的载玻片蘸取细胞 ,干燥后用多聚甲醛固定 ,硝酸银染色。光镜下找到要观察的细胞 ,然后转移到 50目铜网上。电镜下既可看到整个SC核型 ,也可观察同源染色体的联会过程、SC的超微结构及各种类型的SC畸变。该技术可广泛应用于细胞遗传学、遗传毒理学。An improved technique of surface spreading and silver staining for synaptonemal complex preparation was described. Remove mice testicular tissues and make a cell suspension. Place the cells on a surface of 0 4% KCl aqueous solution to spread the SCs by surface tension. Pick up the cells on a formvar covered glass slide. After drying and paraformal dehyde fixation, the synaptonemal complex was stained with silver and observed by light microscopy. Transfer the SCs on formvar to grids for observation in the electron microscope. This technique offers a powerful tool for genome analysis. The ultrastructures of SCs, complex patterns of synapsis and various types of SCs aberrations were clearly observed. It could be widely used in the cytogenetics, genetic toxicology, the reproductive biology etc.
