FAK和整合蛋白α5、β1亚基在胃癌中表达的比较及其临床意义

摘　　要：目的　比较粘着斑激酶 (FAK)和整合蛋白 (integrin)α5、β1亚基在胃癌及癌旁组织中的表达与肿瘤细胞分化、浸润、转移等生物学行为的关系及相互关系。方法　用免疫组织化学亲合素生物素法 (ABC)检测 75例胃癌及癌旁组织石蜡标本FAK和整合蛋白α5、β1亚基蛋白的表达水平。结果　FAK、整合蛋白α5亚基和整合蛋白 β1亚基的表达为 或 分别是胃癌组织 5 7%、2 9%和 2 5 % ,癌旁组织 4%、76%和 84% ;低分化胃癌组织 68%、18%和 16% ,高中分化组织 42 %、45 %和 39% ;有淋巴结转移的组织 77%、31%和 31% ,无转移的组织 36%、2 8%和 19% ;浸润程度较深的组织 68%、2 9%和 2 5 % ,浸润程度较浅的组织 19%、31%和 2 5 %。结论　FAK较整合蛋白α5、β1亚基可以更有效地衡量胃癌的发生与发展 ,FAK的表达量可作为胃癌发生、发展过程中一种较有价值的病理指标。FAK可能既是一种转化相关酶又是一种演变相关酶。Objective To compare the ex pression of focal adhesion kinase (FAK) and integrin α5, β1 subunits in gastri c carcinoma and non-cancerous tissue and study their relations with clinicopath alogical data of the tumor. Methods The expression leve ls of FAK and integrin α5, β1 subunits was detected in 75 gastric carcinomas b y using an immunohistochemical technique. Results The po sitive rate for expression ( or ) of FAK, integrin α5, β1 was 57%, 29% an d 25% in gastric carcinoma tissues, 4%, 76% and 84% in unaffected margin tissues , 68%, 18% and 16% in poor differentiated tissues, 42%, 45% and 39% in well diff erentiated tissues; 77%, 31% and 31% in the tissues with lymph node metastasis, 36%, 28% and 19% in the tissues without lymph node metastasis; 68%, 29% and 25% in the deep infiltrated tissues, 19%, 31% and 25% in the low infiltrated tis sues.Conclusion FAK is a more significant indicator tha n integrin α5, β1 subunit in evaluating carcinogenesis and the progression of the carcinoma. The level of FAK expression might be used as a valuable marker fo r the carcinogenesis and the progression of the carcinoma. The FAK might be not only a transformation linked enzyme but also a progression linked enzyme.
