摘　　要：目的:研究三硫化二砷(As2S3)能否诱导急性粒细胞白血病细胞株(HL60)凋亡与坏死。方法:应用细胞生长测定、形态学观察及流式细胞仪检测等多种方法,在体外研究As2S3对HL60细胞凋亡和增殖抑制的作用。结果:2.5μmol/LAs2S3作用18h后诱导HL60小部分细胞凋亡和大部分细胞坏死7.5μmol/LAs2S3作用8h后可引起HL60细胞凋亡。结论:As2S3可诱导HL60细胞凋亡与坏死,有必要进一步探索其对急性粒细胞白血病治疗的价值。Objective: To investigate whether As2S3 will induce apoptosis and necrosis of human leukemia cell line HL 60. Methods: Acute leukemia cell line HL 60 was used as in vitro model. HL 60 cell line was treated with As2S3 at different concentrations and cell growth was analyzed by trypan blue exclusion. Apoptosis was assessed by cell morphology and flow cytometry. Results: 2.5 μmol/L of As2S3 could induce most of cell HL 60 necrosis and a small part of cell Apoptosis within 18 h,7.5 μmol/L As2S3 could induce apoptosis of human leukemia cell line HL 60 within 8 hours. Conclusion: It is necessary to evaluate the therapeutic value of As2S3 on acute myloid leukemia further. Since it can induce apoptosis and necrosis of HL 60 cell.
