摘 要:目的:观察42℃高温联合α干扰素αinterferonαIFN体外杀伤HL60细胞的效果。方法:HL60细胞经42℃高温处理后加入不同浓度αIFN体外培养,计算清除率。结果:42℃高温或100u/mlαIFN单独应用60min,体外净化骨髓时分别能清除大约89%或62%的白血病细胞。42℃高温联合αIFN能协同灭活HL60细胞,可清除98%以上的白血病细胞。两者联用的最适条件是42℃加αIFN100u/ml作用60min。对粒单系集落形成单位(colonyformingunitgranulocyte/macrophageCFUGM)的形成却没有协同抑制作用且减轻了高温对CFUGM的抑制作用CFUGM的抑制率下降为31.3%与单独高温净化时的47.8%相比有显著差异。结论:42℃高温联合αIFN可以协同杀灭体外的HL60细胞。Objective: To evaluate the effect of α interferon(α IFN) combined with hyperthermia on purging HL 60 cell in vitro. Methods: HL 60 cells were cultured with various concentration of α IFN after hyperthermia and calculated for clearance rate. Results: When bone marrow was purged in vitro with hyperthermia or α IFN 100 u/ml alone, about 89% or 62% of the leukemic cells could be eliminated respectively, combination of α IFN and hyperthermia showed a synergic elimination of HL 60 cells with a clearance rate of above 98%. The best conditions for combination were: temperature kept at 42℃ for 60 minutes with α IFN 100 u/ml. No synergic inhibition of the normal bone marrow colony forming unit granulocyte/macrophage (CFU GM) was observed, while the inhibitory rate of the growth of CFU GM was decreased to only 31.3% which was much lower than that (47.8%) caused by hyperthermia(P< 0.05). Conclusion: Hyperthermia combined with α IFN can kill HL 60 cell synergically in vitro.
