醒脑益智方对SAM-P/10老化痴呆鼠海马超微结构的影响

摘　　要：目的：探讨醒脑益智方治疗老年痴呆作用的形态学机制。方法：采用透射电镜技术，系统观察中药醒脑益智方对ＳＡＭ－Ｐ／１０老化痴呆鼠海马超微结构的影响。结果：治疗各组对海马超微结构均有明显改善，表现在：脂褐素数量、体积均明显减小，溶酶体也明显少于Ｐ／１０空白组；高尔基复合体、粗面内质网等接近于正常Ｒ／１品系状态。多聚核糖体数目增多，预防方表现为线粒体基本正常，单位膜清晰，嵴无断裂。其它海马神经元部分都有明显改善，但都以预防方和１号方效果最明显。结论：海马神经元病变符合中医“髓海不足”、“脑髓消减”的改变，而脂褐素形成增加、线粒体肿胀变性，则为“痰浊”等实邪病因提供了客观依据。用补肾健脑、化瘀开窍方早期预防给药及治疗老年痴呆，有明显作用，并优于西药组。To explore the morphology mechanism of Xinnao Yizhi prescription treating senile dementia.Methods:To observe the effect of Xinnao Yizhi prescription on the hippocampal ultrastructure of SAM-P/10 by transmission electron microscope. Results: Obvious improvement of all groups' hippocampal ultrastructure after being treated is following: the number, volume of lipofuscin reduced obviously, lysosome is lower than that of control group. Golgi apparatus, rough endoplasmic retiulum are restored to state of SAM-R/1. The number of polyribosome increased. The state of preventive prescription is following: mitochondria approach normal level, unit membrane is lucidity cristae has no split.The other groups is not so much as it. The improvement of hippocampal nerve cell of preventive prescription and prescription I has the best effect. Conclusion: The disease of hippocampal never cell match with 'medullary sea deficiency' of TCM. The increasing of lipofuscin, degeneration of mitochondria provided objective bases for the pathogeny of 'phlegm turbidity . The effect of the prescription of reinforcing kidney, strengthening brain, resolving stasis and restoring a clear head are better than that of western medicine, which prove it too.
