摘 要:目的 观察大鼠局灶性脑缺血后神经元内细胞周期蛋白D1(cyclinD1)表达与神经细胞凋亡的关系以及丹参的影响。方法 采用线栓法大鼠大脑中动脉持续阻塞模型 ,应用免疫组织化学和TUNEL染色方法观察假手术组和缺血后不同时间阳性染色神经细胞的数量和分布情况及丹参的影响。结果 cyclinD1蛋白表达自缺血后 6h开始出现 ,缺血后 48h达高峰 ;丹参组各相应时间点cyclinD1蛋白表达明显减少 (P <0 .0 1)。凋亡细胞自缺血后 12h开始出现 ,缺血后 72h达高峰 ;丹参组各相应时间点凋亡细胞表达明显减少 (P <0 .0 1)。相邻切片可见cyclinD1蛋白表达和凋亡细胞染色区域基本相同。结论 脑缺血后cyclinD1的异常表达可能诱发了缺血神经细胞的凋亡 。Objective To investigate the effects of radix salviae miltiorrhizae(RSM) on expression of cyclin D1 protein in neurons and neuronal apoptosis at different intervals following permanent focal cerebral ischemia in rats.Methods A model of the middle cerebral artery occlusion (MCAO) in rats was established with intraluminal filament occlusion.The RSM group of rats was treated with RSM 30 minutes before and 2,3 days after the operation.The immunohistochemistry and TUNEL staining were used respectively to observe the distribution and quantities of cyclin D1 protein and neuronal apoptosis in the brain tissues following the sham operation and permanent MCAO at 3 h,6 h,12 h,24 h,48 h,72 h and 7 d respectively.Results A significant increase in cyclin D1 immunostaining and TUNEL positive staining was detected at 6 h and 12 h after MCAO in the neurons of the ischemia penumbra,and was most marked at 48 h and 72 h.The differences between these groups were significant (P<0.01).Conclusion The induction of cyclin D1 protein expression at the ischemia penumbra indicated that it might be involved in the mechanism of neuronal apoptosis.RSM might reduce the neuronal apoptosis through down -regulating cyclin D1 expression.
