摘 要:建立了可用于诊断苯丙酮酸尿症的5种尿中微量芳香酸的毛细管电泳分析新方法。应用硼砂-胆酸钠(均为25 mmol/L,pH=10.0)电泳介质体系,可将5种芳香酸在15 min内达到全分离;最低检测限达1.5×10-9 mol;采用Sep-Pak C18微柱进行尿样前处理,有效地除去了尿蛋白对分离的干扰,可作为苯丙酮酸尿症的筛查方法。Micro-aromatic acids play an important role in the diagnosis of organic acidurias. In this study, a simple and rapid capillary electrophoresis ( CE) method for the analysis of micro-aromatic acids in urine had been developed, the electrophoretic behaviour of these aromatic acids had been discussed in detail. The electrophoresis was carried out in a 70 cm long, 50 mum ID fused-silica capillary, filled with 25 mmol/L borax buffer (pH = 10.0) containing 25 mmol/L sodium cholate as the electroosmotic flow modifier at a protential of 16 kV. The detection was at 214 mn. Under this condition, five phenylketonuria (PKU) related aromatic acids had been separated within 15 min, the detection limit was approximately 1.5 x 10(-9) mol for 4-hydroxyphenylacetic acid.
