摘  要:采用PCR-SSCP分析技术,检测40例大肠癌癌组织中抑癌基因APC第11外显子、MCC第10外显 子和癌基因 K-ras第 1外显子的变异情况,并在 20例大肠癌患者粪便中进行三种基因的检测。结果:APC第 11 外显子和 MCC第10外显子在 A/B期变异率明显高于 C/D期,K-ras第 1外显子突变率在粘液癌中明显高于腺 癌,且随分化程度降低有降低的趋势;在粪便中均检测到与肿瘤组织中三种基因相应的变异。认为检测粪便脱落 细胞基因变异可用于大肠癌早期诊断,APC/MC和 K-ras可用于估计大肠癌预后。The changes of exon 11 of APC (adenomatous polyposis coli) gene, exon 10 of MCC(mutated in colorectal carcinoma)gene and exon 1 of K-ras (Kirsten rat sarcoma)gene were separately assayed with PCRSSCP(polymerase chain reaction-single strand conformation polymorphism) technique in 40 samples of colorectal carcinoma and 20 samples from stools of patients with colorectal carcinoma. Results showed the alterative rate of exon 11 of APC and exon 10 of MCC were significantly higher in stage A/B than in stage C/D. The mutation rate of exon 1 of K-ras were significantly higher in mucinous carcinoma than in adenocarcinoma, and the rate had a decreasing tendency with the differentiation decreased. It is thought that detection of APC/MCC and K-ras genes might be used in early diagnosis and prognosis of colorectal carcinoma.
