摘 要:目的 :探讨IL - 1β在咬合力影响牙周组织改建中的分子机制。方法 :采用Wistar大鼠建立正常咬合力、咬合力丧失和增强的动物模型。用HE染色和免疫组化的方法 ,观察不同咬合力状态下牙周组织的形态学改变以及IL - 1β在牙周膜成纤维细胞 (PDLC)和牙槽骨成骨细胞中表达的动态变化。 结果 :咬合力丧失导致牙周膜结构紊乱、牙槽骨吸收 ,IL - 1β在PDLC和成骨细胞中的表达 ,较正常咬合力时明显增强。咬合力增强引起牙周膜结构致密、宽度增加 ,牙槽骨有明显的新骨形成 ,IL - 1β在PDLC和成骨细胞中的表达也较正常时明显增强。结论 :咬合力丧失和增强均诱导IL - 1β在牙周组织PDLC和成骨细胞中的表达 ,较正常咬合力时明显增强 ,提示IL -AIM: To study the expression of IL-1β in periodontal ligament fibroblasts (PDLC) and osteoblasts in order to explain the molecular mechanism of the periodontium remodeling under biting force. METHODS: A rat model of different biting force was established by extracting the left maxillary molars, so that the left mandibular molar area was used as the biting force lost model and the right mandibular molar area as the biting force increased model. The untreated rats' mandibular molar area used as normal biting force model. The prepared tissue specimens were processed to study the change in histologic morphology with HE staining, and the expression of IL-1β in PDLC and osteoblasts with immunohistochemistry (IHC) technique. RESULTS: In the group of lost biting force, derangement of periodontal ligament and resorption of alveolar bone were observed in histologic study. The expression of IL-1β in PDLC and osteoblasts enhanced apparently than in the group of normal biting force with IHC technique . In the group of increased biting force, the same result was observed with IHC, but increase of periodontal ligament space and more dense of the ligament, resorption and formation of alveolar bone, were observed under histologic study. CONCLUSION: The results of both lost and increased biting force enhanced the expression of IL-1β in PDLC and osteoblats suggested that the IL-1β may play important roles in the process of periodontium remodeling initiated by different biting force.
