小剂量金喜素与顺铂、VP16及5-FU对人卵巢癌细胞株生长抑制作用的实验研究

摘　　要：目的 :探讨小剂量国产盐酸拓扑替康 (金喜素 )与顺铂 (cDDP)、足叶乙甙(VP16 )及 5 -氟尿嘧啶 (5 -FU)对人卵巢癌细胞株 3AO细胞生长抑制的作用。方法 :采用MTT法观察上述药物单用及联合使用对 3AO细胞生长的影响 ,并评价药物的联合作用。结果 :相当于临床用药浓度的金喜素 (80 0ng/ml)及其半量浓度 (40 0ng/ml)与cDDP、5 -FU或VP16联合应用对 3AO细胞具有显著的协同生长抑制作用 ,较各药单用组抑制率显著提高。半量金喜素联用即可达到全量联用时的抑制效果。结论 :小剂量金喜素与cDDP、5-FU或VP16具有协同抑制 3AO细胞生长的作用 。Objective:To study the growth-inhibition effect of low dose Topotecan Hydrochloride (Jin Xi Su) with the combination of cisplatin,5-fluorouracil and etoposide.Methods:The effects of individual of combined drugs on 3AO cells were observed by MTT assay,coefficient of drug interaction was used to analysis the synergistic effect of these drugs.Results:800ng/ml Jin Xi Su which is considerably equal to clinical effective concentration and its half-dose concentration 400ng/ml could significantly enhanced the anti-proliferation effects of cDDP,5-FU or VP16 when combined with them,and their inhibitory rates were significantly higher than those in single drug groups.Half-dose Jin Xi Su Combination group could reach the similar cell growth inhibitory rate of that in entire-dose group. Conclusions:Jin Xi Su in small dosage can synergistically inhibit the proliferation of 3AO with the combination of cDDP,5-FU or VP16,which can give reference for low dosage use of Topotecan in chemotherapy.
