胃癌幽门螺杆菌感染与微卫星不稳定性的研究

摘  要:目的 通过研究胃癌幽门螺杆菌 (Hp)感染与微卫星不稳定性 (MSI)的关系 ,探讨幽门螺杆菌在胃癌发生中的作用。方法 采用PCR为基础的方法对 41例胃癌组织中HpCagA基因以及 4个微卫星位点BAT2 6 ,TP5 3,D2s12 3,D2s119进行检测 ,分析Hp感染与微卫星不稳定性之间的关系。 结果 胃癌Hp和CagA的阳性率分别为 6 3.4% (2 6 / 41) ,6 9.2 % (18/ 2 6 )。BAT2 6 ,TP5 3,D2s12 3,D2s119位点MSI的检出率分别为2 1.9% (9/ 41) ,2 9.3% (12 / 41) ,2 9.3% (12 / 41)和 14.6 % (6 / 41)。Hp阳性组各位点MSI检出率与Hp阴性组比较无显著差异 (P >0 .0 5 )。CagA+ Hp组与CagA-Hp组各位点MSI检出率差异亦无显著性 (P >0 .0 5 )。 结论 Hp感染与MSI无显著相关 。Objective To evaluate the role of Helicobacter pylori (Hp) on the development of gastric cancer. Methods The CagA gene of Hp and microsatellite instability (MSI) at loci BAT26, TP53, D2s123 and D2s119 were examined with PCR based technique. The relationship of Hp infection with MSI was analyzed. Results The positive rate of HP and CagA gene was 63.4% (26/41) and 69.2% (18/26) respectively. MSI was detected in 21.9% (9/41) at BAT26,29.3%(12/41) at TP53,29.3%(12/41) at D2s123, and 14.6%(6/41) at D2s199. No significant difference was found in the frequency of MSI between Hp positive and Hp negative cases ( P >0.05). No significant difference was also found in the frequency of MSI between CagA + group and CagA - group ( P >0.05). Conclusion Hp infection shows no association with MSI and may not exert its effect on the development of gastric cancer through MSI.
