人脊索瘤细胞的体外侵袭性

摘　　要：目的　观察人脊索瘤细胞在体外侵袭性的大小 ,探讨与肿瘤侵袭性密切相关的尿激酶型纤溶酶原激活物 (uroki nase typeplasminogenactivator,uPA)在侵袭中的作用 .方法　利用体外球体侵袭模型观察人脊索瘤细胞的侵袭特性和侵袭过程 ;应用免疫组化SABC法检测脊索瘤细胞中uPA的表达情况 ,并且观察用uPA抗体后肿瘤细胞侵袭性的变化 .结果　肿瘤细胞球体在接触预培养的鸡胚心肌块 (precultureheartfragment,PHF)后 3d即包绕PHF ,并有部分侵袭 ,接触培养 5d ,每个PHF即有一半左右被侵袭 ,10d左右 ,PHF即完全被侵袭 .免疫组化染色 ,阳性细胞占 6 3% ,为uPA高表达 .在培养液中加入uPA抗体后 ,肿瘤细胞侵袭性减低 .结论　人脊索瘤细胞具有一定的侵袭性 ;该细胞uPA高表达 ;AIM To evaluate the invasion of human chordoma cell in vitro , and the effects of urokinase type plasminogen activator on tumour invasion. METHODS The invasive capacity and the invasive process of chordoma cells were observed with invasion model of tumour cell spheroid aggregate in vitro ; Applying streptavidin biotin complex (SABC) immunohistochemical technique, the expression of uPA in human chordoma cell line was studied. RESULTS 3 d after PHFs confronted cultured with tumour cell spheroid aggregates, they were sphered and invaded. 5 d after confronting cultured, about the half of PHF was invaded and 10 days after, most of PHFs were completely invaded. uPA was expressed in human chordoma cell, the positive rate was 63%. The invasion of chordoma cell was decreased when the monoclonal antibodies of uPA were added in fluid medium. CONCLUSION Human chordoma cell line has a high capacity of invasion in vitro , and has high uPA expression. The antibody of uPA may decrease the invasion of chordoma cell line.
