摘  要:目的 探讨诱生型一氧化氮合酶 (iNOS)及其催化生成的一氧化氮 (NO)在肺癌发病机制中的作用。方法 采用逆转录 -聚合酶链反应 (RT -PCR)技术检测 40例不同病理类型、不同临床分期肺癌患者癌组织中iNOSmRNA的表达情况 ;同时分别以ELISA法、硝酸还原酶法测定相应血清中肿瘤坏死因子 -α(TNF -α)及NO的浓度。设立对照组比较。结果 肺癌组iNOSmRNA的阳性表达率为 72 .5 0 % ,较对照组 (11.76 % )明显增高 (P <0 .0 1)。其中鳞癌、腺癌、小细胞癌的iNOS表达率分别为 6 0 .11%、82 .35 %、80 .0 0 % ,不同病理类型间无显著性差异 (P >0 .0 5 ) ;但早期肺癌 (Ⅰ~Ⅱ期 )iNOS阳性率 (87.5 0 % )则高于晚期肺癌 (Ⅲ~Ⅳ期 )的阳性率 (5 0 .0 0 % ) (P<0 .0 5 )。肺癌组血清NO和TNF -α水平较对照组显著升高 (P<0 .0 1)。结论 肺癌组织在TNF -α刺激下可表达iNOS ,iNOS通过催化合成NO ,在分子水平对肺癌的发生。Objective To explore the role of inducible nitric oxide synthase (iNOS) and nitric oxide (NO) in the development of lung cancer.?Methods The expression of iNOS was detected by reverse transcription-polymerase chain reaction (RT-PCR) in 40 cases of lung cancer of different pathological types at different clinical stages.The contents of NO and tumor necrosis factor-α(TNF-α) in serum were determined by spectrophotometry and ELISA respectively.?Results ①The target gene of iNOS was detected in 29 of 40 tumor tissues (72.50%) and in 2 of the 17 normal tissues ( 11.76% ) ( P <0.01 ). The frequencies of gene expression of iNOS were 61.11% in squamous cell carcinoma, 82.35% in adenocarcinoma and 80.00% in small-cell lung cancer respectively. No significant difference was found between the different histologic types ( P > 0.05 ).But the frequency of iNOS expression was higher in the early clinical stages of lung cancer (Ⅰ-Ⅱ) than in the advanced ones (Ⅲ-Ⅳ) ( P <0.05). ② The concentrations of serum NO and TNF-αin lung cancer patients were obviously higher than those in controls ( P <0.01).?Conclusion The iNOS gene may be expressed in human lung cancer under the effect of cytokine TNF-α. The iNOS may be involved in the development of lung cancer at a molecular level through the thynthsis of NO catalyzed by it.
