不同种类抗角蛋白抗体的应用比较

摘　　要：抗角蛋白抗体是鉴别上皮性肿瘤的有效标记物之一,由于角蛋白分子量多种多样,故形成之抗体亦不完全相同,其免疫反应亦异。本文应用8种不同种类之抗角蛋白抗体(其中5种系北京生研所提供)对87例各种类型之肿瘤比较了它们在免疫组化反应上的差异。结果显示,多克隆抗体k-D反应谱系较广,且对低分化鳞癌的反应亦较好,但非特异性染色较重易造成判断上的困难。单克隆抗角蛋白抗体的特异性较强,非特异性染色轻。但反应谱较窄。其中HK2对腺癌的敏感性较高,如将HK2、HK5按1∶1比例混合使用则更可提高其对腺癌的反应程度,同时还相应提高了对鳞癌的反应能力。Anti-keratin antibody is an useful tumor marker to differentiateepithelium and non-epithelium tumors. As the molecular weightsof various keratins are different so many kinds of antibodies areprovided. We used 5 monclonal antibodies (HK_2, HK_5, HK_(23), K_9,K_(12)) and 3 other commercial anti keratin antibodies (AE3 AE8 K-D) to compare their immuno-reaction in 87 cases of differenttypes of tumors. The results show that the polyclonal antiserumK-D has a wide spectrum reaction, and it is especially useful to thesquamous cell carcinoma even in the poorly differentiation typetu-mors as the reaction is still present, but the non specific stainis too heavy. The HK serial monoclonal antibodies has their ownspecific reaction spectrum and the background is clear but the rangeof reaction is narrow. It is found that HK_2 has a high affinity foradenocarcinoma, and the mixture antibodies (HK_2 ad. HK_5, 1:1) couldexpand their ability and were better than K-D.
