摘  要:目的 观察博莱霉素 (bleomycin ,BLM)所致小鼠肺损伤后纤维化时广谱钙粘附素 (pan cadherin,Pan cd )及 β连环素 (β catenin ,β cat)表达的动态变化 ,探讨钙粘附素、连环素表达与肺纤维化发生发展的关系。方法 小鼠气管内注入BLM复制肺纤维化模型。分期处死动物 ,以抗Pan cd及抗β cat抗体行肺组织免疫组织化学染色。 结果 肺纤维化时 ,Ⅰ型肺泡细胞Pan cd表达下调 ,Ⅱ型肺泡细胞被新诱导表达Pan cd。随后 ,细支气管上皮Pan cd与 β cat的表达均下调 ,并呈现出下调 恢复 下调的动态变化 ,同时伴Pan cd表达由胞膜型转变为胞浆型 ,且呈现出胞浆型 胞膜型 胞浆型表达的动态变化。肺泡巨噬细胞 (AM)及肺间质细胞Pan cd表达增加。结论 Pan cd、βObjective To investigate the changes of pancadherin(Pan cd) and β catenin(β cat) expression in pulmonary tissue of mice with bleomycin (BLM) induced pulmonary injury and explore the relation between the Pan cd/β cat expression and pulmonary fibrosis. Method A single dose of BLM was intratracheally injected to induce pulmonary fibrosis of mice. Animals were killed and immunohistochemical methods were used with polyclonal anti Pan cd and anti β cat antiserum. Results The expression of Pan cd in typeⅠalveolar cells was down regulation. The expression of Pan cd in typeⅡalveolar cells was induced. The expression of Pan cd and β cat in small airway epithelial cells were all down regulation and were repeated with the way of down regulation reparation down regulation. The expression of Pan cd on membrance in small airway epithelial cells decreased,whereas the cytoplasmic expression increased and was repeatedly alterated between cytoplasma with membrance. The expression of Pan cd in alveolar macrophage and interstitial cells increased.Conclusion The abnomal` expression of cadherin and β cat may play a role in the pathogenesis of pulmonary fibrosis after pulmonary injury.
