摘  要:目的:探讨端粒酶作为胶质瘤标志物,在肿瘤良恶性鉴别、恶性程度评估方面的意义。方法:利用TRAP-ELISA方法定量检测47例胶质瘤中端粒酶的活性,并结合肿瘤的分类和患者的临床资料进行分析。另取10例正常脑组织标本作为对照。结果:47例胶质瘤中,高分化胶质瘤(星形细胞瘤Ⅰ~Ⅱ级、少支胶质细胞瘤和室管膜瘤)24例,端粒酶阳性2例(8.3%);低分化胶质瘤(星形细胞瘤Ⅲ~Ⅳ级和间变性室管膜瘤)23例,端粒酶阳性20例(87.0%)两组之间端粒酶阳性率有显著差异(P=0.001);另外,两组之间端粒酶活性水平定量也有显著差异(P=0.022)。端粒酶阳性组和端粒酶阴性组胶质瘤患者在性别、年龄方面无显著差异(P值分别为0.704和0.905)。10例正常脑组织中未检测到端粒酶活性。结论:端粒酶活性的激活在胶质瘤中是常见的现象。端粒酶作为肿瘤标志物,在脑胶质瘤的良恶性鉴别、恶性程度评估方面具有重要的意义。Objective: To investigate the telomerase activity in gliomas and its significance in classification and gradation of the tumors. Method: Telomerse ploymerase enzyme linked immunoassay (TRAP-ELISA) was used to detect the telomerase activity in 47 patients with gliomas. Another 10 normal brain tissues were employed as the control group. Result: In 47 gliomas, 2 of 24(8.3%) classical low grade gliomas (astrocytoma Ⅰ-Ⅱ, oligodendroglioma, and ependymoma) were detected to be telomerase positive; 20 of 23 (87%) classical high-grade gliomas (astrocytoma Ⅲ-Ⅳ and anaplastic ependymoma) were positive in telomerase expression. The difference of the positive rate between these two groups was significant (P=0.001). The quantitative difference of telomerase activity were also significant (P=0.022). There was no difference of patient′s gender and age between telomerase-positive group and the telpmerase-negative group (P values were 0.704 and 0.905 respectively). Telomerase activity was not detected in 10 normal brain tissues. Conclusion: Reactivation of telomerase activity may be a frequent event in malignant gliomas. As a tumor maker of gliomas, telomerase has significant importance in differentiating begin and malignant, malignancy evaluation of gliomas.
