摘 要:目的 观察钙离子拮抗剂尼莫地平 ( NMD)与化疗药物卡氮介 ( BCNU)及威猛 ( Vm-2 6)联合使用对脑胶质瘤细胞化疗耐药性的影响 ,为临床上脑胶质瘤的合理化疗提供理论依据。方法 通过体外培养的 C6胶质瘤细胞 ,采用 MTT法及 3 H-Td R掺入法 ,比较 NMD与 BCNU、Vm-2 6单独使用、联合使用对肿瘤细胞增殖率的影响。结果 NMD分别与 BCNU、Vm-2 6联合使用对肿瘤细胞的抑制率明显高于三者单独使用的抑瘤效果 ( P<0 .0 1 ) ,而三者联合使用对肿瘤细胞的抑制率可达 97.0 4 % ,也明显高于 NMD与 BCNU或 Vm-2 6联合使用的抑瘤率 ( P <0 .0 1 )。同时 ,联合化疗时 BCNU、Vm-2 6的用药浓度仅为单独使用的 0 .1~ 0 .0 1。结论 NMD可以明显降低胶质瘤细胞对化疗药物 BCNU和 Vm-2 6的耐药性 ,而且可以降低化疗药物的用药浓度。Objective To study the effect of calcium antagonist nimodipine(NMD) combined with chemotherapy drugs carmustine(BCNU) and teniposide (Vm 26) on chemoresistance in malignant glioma cells and provide rational chemotherapy method in clinical.Methods MTT method and 3H TdR incorporation assay were applied in single layer cultured C 6 glioma cells to observe the cell proliferation rates when NMD, BCNU and Vm 26 were singly or combinatively used respectively. Results The inhibitory rates of NMD combined with BCNU and Vm 26 respectively were higher than those when they were singly used (P<0.01). The inhibitory rates of NMD combined with BCNU and Vm 26 together(97.04%) were higher than the effects of NMD combined with BCNU or Vm 26 (P<0.01). The anticancer drug concentration in combination chemotherapy were 10 to 100 times lower than those of single chemotherapy.Conclusion NMD can significantly decrease the chemoresistance of BCNU and Vm 26 in malignant glioma cells, at the same time , it can decrease the chemotherapy drug concentration as well.
