用免疫组化和原位杂交方法检测和分析人骨肉瘤组织中Ⅰ、Ⅱ 和Ⅲ型胶原蛋白和mRNA表达

摘　　要：目的　观察骨肉瘤组织 、 和 型胶原蛋白和 m RNA的表达及其与骨肉瘤分型和分化的关系。方法　免疫组化 LSAB法检测胶原蛋白 ,原位杂交检测胶原 m RNA的表达 ,用图像分析系统进行分析和半定量测定。结果　骨肉瘤组织不仅表达 型胶原蛋白 (阳性率 87% ,灰度值1 71 .99± 1 4.74) ,并出现了 型和 型胶原蛋白 (阳性率分别为 30 .4%和 43.4% ,灰度值分别为1 5 3.0 7± 1 8.82和 1 68.2 9± 1 8.36) ;软骨母细胞型和混合型骨肉瘤 型胶原的表达高于其它型 ,差异有显著性 ( P<0 .0 1 ) ;高中低分化的三组骨肉瘤间 , 型胶原表达差异有显著性 ( P<0 .0 1 ) ,分化越低表达越少。 、 型胶原 m RNA表达与蛋白表达所得结论基本一致。结论　 型胶原可作为软骨母细胞型和混合型骨肉瘤的分型参考指标 ,Objective To investigate expression of type Ⅰ?Ⅱ and Ⅲ collagen in human osteosarcoma and their relationship with classification and differentiation of osteosarcoma.Methods The protein and mRNA of type Ⅰ?Ⅱ and Ⅲ collagen in 46 cases of osteosarcoma were detected using immunohistochemistry (LSAB) and in situ hybridization;EMAL 200 image software was used to analyse staining result.Results Besides type Ⅰ collagen (positive rate 87%,grey value 171.99±14.74),there were also expression of type Ⅱ and Ⅲ collagen in osteosarcoma(positive rate are respectively 30.43% and 43.5%,grey value respectively 153.07±18.82 and 168.29±18.36);Type Ⅱ collagen stain was significantly stronger in chondroblastic and mixed osteosarcomas than other type.Among three groups of osteosarcomas of high,moderate and low degree,the difference of type Ⅰ collagen was statistically significant,the lower osteosarcoma differentiated,the weaker expression of type Ⅰ collagen was.The ISH of mRNA revealed analogous change to protein.Conclusion Type Ⅱ collagen can be utilized as one of referring indicators to identify chondroblastic and mixed osteosarcomsa;Type Ⅰ collagen can be used as one of markers to determine the differentiation and malignancy of osteosarcoma.
