慢性粒细胞性白血病抗原负载树突状细胞激发的抗白血病作用

摘　　要：目的　研究慢性粒细胞性白血病 (CML)细胞冻融抗原 (CLA)负载的树突状细胞 (DC)对特异性抗白血病T细胞的作用。方法　将CML患者外周血单个核细胞 (PBMNC)来源的DC在体外用CLA负载 ,再与CML患者的经细胞因子诱导的杀伤细胞 (CIK)共同培养 ,应用乳酸脱氢酶 (LDH)释放法观察其对自身CML细胞杀伤活性 (A组 ) ,与未负载的DC +CIK(B组 )、CIK(C组 )及CIK +CLA(D组 )进行比较。结果　效靶比为 10∶1条件下 ,A、B、C、D 4组的杀伤活性分别为 (6 3 6 9± 8 35 ) %、(4 5 0 2± 5 49) %、(2 7 2 8± 4 6 4) %、(2 9 6 3± 5 71) %。A组比B组杀伤活性强 (P <0 0 1) ;C组与D组比无显著性差异。Objective To investigate the immunostimulatory effect of dendritic cells(DC) pulsed with chronic myelogenous leukemic lysate antigen (CLA).Methods Dendritic cells from chronic myelogenous leukemia(CML)patients were pulsed by CLA,and then cocultured with cytokine-induced killer cells(CIK) from CML patients(group A).The cytotoxic activity was measured by using a lactate dehydrogenase release assay,and compared with DC+CIK(group B),CIK(group C) and CIK+CLA(group D).Results The cytotoxic activity of four groups against autologous CML cells was (63 69±8 35)%,(45 02±5 49)%,(27 28±4 64)% and (29 63±5 71)%,respectively.There was significant differences between group A and group B or between group B and group C.Conclusion Coculture of CIK cells with DC led to a significant increase in cytotoxic activity.The cytotoxic activity could be further increased by using DC pulsed with CLA which can increase the cytotoxic activity of immunologic effector cells against CML cells.DC pulsed with CLA may have an important effect on immunotherapy protocols for patients with chronic myelogenous leukemia.
