雌三醇阴道栓剂的药效学及局部毒性的研究

摘　　要：目的 :研究雌三醇 (E3)阴道栓剂的药效学及局部毒性。方法 :用去卵巢小鼠 ,局部给E3阴道栓剂后 ,观察对阴道角化细胞及子宫重量的影响 ;大鼠局部给E3阴道栓剂后 ,观察对阴道的刺激性及毒性。结果 :按 12 5、2 5、5 0 μg·( 10g) 1体重为去卵巢小鼠阴道内一次给药后 ,4 8～ 72h ,小鼠阴道上皮角化细胞动物阳性率为 75 %～ 10 0 % ,阴道角化细胞等级积分总和分别为 2 8～ 4 8和 2 4～ 4 2 ,子宫除 12 .5 μg·( 10g) 1剂量组外均有显著增重作用。大鼠局部用药 ,无刺激性及全身毒性。结论 :试验证实E3阴道栓剂安全 。OBJECTIVE:To study on pharmacodynamics and local toxicity of estriol pessary.METHODS:We have observed the effects of estriol pessary on vaginal cells and uterus weight of mice whose ovaries were artificially moved and local toxicity in normal rats. RESULTS:After taking a single local dose 12.5,25,50 μg·(10 g) 1 estriol pessary were increased 75%～100% in positive reaction mice on hornification cells,equal stage accumulate minute sum were 24 48,24 42 in 48 72 hours, respectively. Uterus weight was increased except 12.5 μg·(10 g) 1No local stimulation and systemic toxicity were observed with local application.CONCLUSION:The results show that estriol pessary is safe and effective.
