摘 要:目的 :建立测定高原雪灵芝中阿魏酸含量的高效液相色谱法。方法 :采用碱液浸取、加酸沉淀后再用乙醚提取雪灵芝中的阿魏酸 ;色谱柱为 μBondapakODS柱 ,流动相为甲醇 水 冰醋酸 ( 4 0∶60∶3 ) ,检测波长 3 0 5nm。 结果 :雪灵芝中的阿魏酸与样品中其它成分分离良好 ;阿魏酸在 90~ 80 0 μg·ml-1范围内 ,峰面积与浓度的线性关系良好 (r=0 9997) ;平均回收率为 99 5 9%;日内精密度RSD =0 79%(n =5 ) ,日间精密度RSD =0 86%(n =5 )。结论 :该法操作简便 ,结果准确 。OBJECTIVE:To establish a reversed phase high performance liquid chromatography for determining the content of ferulic acid in Xuelingzhi ( Arenaria Kansuensis Var.ovalipeatala ) on plateau.METHODS:The samples were extracted with 1.2% NaOH solution,then the extract was acidified to pH 4.0 with HCl and extracted with ether,ferulic acid was determined on LC 3A HPLC instrument fitted with a μ Bondapak C 18 column (25 cm× 4.6 mm,10 μm)with detector at 305 nm,and the mobile phase consisted of CH 3OH H 2O CH 3COOH(40∶60∶3),the flow rate was 0.8 ml·ml -1 RESULTS:Ferulic acid and other components in Xuelingzhi can be separeted.The calibration curves were linear( r = 0.9997 )within the range of 90~800 μg·ml -1 for ferulic acid.The average recovery was 99.59% , RSD in a day was 0.79%( n =5)and RSD between days was 0.86%( n =5) at three levels.CONCLUSION:This method is simple and accurate and is suitable to the quality control for Xuelingzhi.
