摘 要:目的 探讨经脑室直接注射胰岛素对全脑缺血后大鼠学习记忆力的影响。方法 以 4 VO法建立大鼠全脑缺血再灌注模型。全脑缺血 2 0min后行再灌注 ,于再灌注即刻经脑室注入 1U胰岛素 ,缺血再灌注后 8周利用三等分Y型迷宫箱测试大鼠的学习记忆功能 ,并对大鼠海马CA1区神经元的病理改变进行观察。结果 经Y型迷宫测试 ,对照组、治疗组及缺血组大鼠达 9/ 10正确反应所需电击数分别为 5 .6±1.2 ,8.6± 1.3,19.6± 2 .6 ;对照组、治疗组及缺血组大鼠海马CA1区神经元计数分别为 176 .5± 10 .6 ,112 .4± 11.7,10 .5± 0 .4。结论 全脑缺血后脑室内注射胰岛素可明显减少大鼠海马CA1区神经元的死亡 ,进而减轻大鼠学习记忆力损害。Objective To investigate the effect of insulin directly given into the ventricle on the changes of memory of rats following global brain ischemia. Methods Fourvessel occlusion was used to establish the model of rat global brain ischemia. 1U insulin was injected into the ventricle immediately after 20min brain ischemia. 8 weeks after reperfusion, the ability of memory of rat was tested by 'Y' maze, and the pathological changes in hippocampus CA 1 region was observed. Results Tested by 'Y' maze, the numbers of electric shock for the criteria (9 right reactions after 10 times electric shock) in control, treated and ischemic groups were 5.6 ± 1.2 , 8.6 ± 1.3 , 19.6 ± 2.6 respectively. The counts of normal neuron in control, treated and ischemic groups were 176.5 ± 10.6 , 112.4 ± 11.7 and 10.5 ± 0.4 respectively. Conclusion Intraventricular administration of insulin following global brain ischemia can reduce death of neurons in hippocampus CA 1 region and attenute the memory deficit in rats following transient global brain ischemia.
