PCNA、CD44v6在原发性肝细胞癌的表达及临床意义

摘　　要：目的探讨 PCNA、CD44v6在肝细胞癌(HCC)组织的表达及其临床病理意义。方法采用 PCNA、CD44v6单克隆抗体,对83例 HCC 标本进行免疫组化染色。结果 PCNA、CIN4v6在 HCC 的阳性率分别为87.95%和53.01%。PCNA 在 Edmondson Ⅰ、Ⅱ、Ⅲ、Ⅳ级的阳性率分别为42.86%、89.47%、92.86%、100%,有显著性差异(P<0.01)。CD44v6在转移组的高表达率为39.29%,明显高于无转移组(10.91%,P<0.01)。CIN4v6在包膜完整组和不完整组的阳性率分别为35.42%和77.14%,有显著性差异(P<0.01)。CD44v6阳性病人的生存期明显短于 CD44v6阴性病人(P<0.05)。PCNA 和 CD44v6在 HCC 的表达呈正相关。结论 PCNA、CD44v6有望成为预测HCC 发生、发展、转移、预后的生物学指标。Objective To investigate expression of PCNA and CD44v6 in human hepatocellular carcinoma(HCC)and explore its clinical significance.Methods Surgically resected specimens of 83 cases of HCC were studied immunohistochemically by using a mAh against PCNA and a mAb against CD44v6.An antigen retrieval method followed by a routine ABC method was employed.Results PC- NA and CD44v6 were detected in 87.95% and 53.01% of HCC specimens,respectively.The positive rates of PCNA detected in Edmondson Ⅰ,Ⅲ,Ⅲ and IV of HCC were 42.86%,89.47%,92.86% and 100%,respectively.There was a significant correlation between PCNA immunoreactivity and his- tological type of the tumor(P<0.01).CD44v6 was detected in 39.29% of HCC with tumor metasta- sis,while only 10.91% of HCC without any metastasis(P<0.01).Furthermore,CD44v6 was detec- ted in 35.42% of HCC with entire envelope,compared with 77.14% of HCC without entire envelope (P<0.01).CD44v6-positive HCC patients showed a significantly shorter survival period after surgi- cal operation than those CD44v6-negative HCC ones(P<0.05).The expression of PCNA was corre- lated to that of CD44v6.Conclusions The expression of PCNA and CD44v6 is correlated with the oc- currence,development or metastasis or HCC,as well as with worse prognosis of patients with HCC. PCNA and CD44v6 are hopefully to be a useful marker for HCC.
