侧耳属一新菌株及其亲本的生化特性

摘  要:对佛罗里达侧耳和姬菇单孢菌丝经杂交获得的-新菌株及其亲本,应用聚丙烯酰胺凝胶不连续垂直平板电泳方法,测定了酯酶、过氧化物酶、醇脱氢酶、乳酸脱氢酶和苹果酸脱氢酶等5种同工酶酶谱;应用薄层等电聚焦电泳方法测定了可溶性蛋白;应用硅胶G薄层层析法鉴定多糖的组成成分。结果表明:新菌株与亲本之一的姬菇,在3种脱氢酶酶谱上甚为相似;新菌株与亲本的酯酶同工酶区带丰富,新菌株多数区带来自亲本,与姬菇相似程度高,但有新区带出现;新菌株的多糖组成成分与佛罗里达侧耳相似;新菌株与亲本的可溶性蛋白无明显区别。推断该新菌株为杂交异核体。This paper presents the analysis of live lsozymes (Est., POD, MDH, LDH and ADH) and soluble proteins of a new strain and its parents (Pleurotus florida and Pleurotus sp.), by use of vertical slab and thin-layer isoelectric focusing in polycrylamide gel. The results show that five zymograms of the new strain are more similar to that of Pleurotus sp. than Pleurotus florida, whereas all of their soluble proteins are similar each other. An analysis of the polysacchride components with silica gel G layer chromatography shows that of the new strain is similar to Plcurotus florida. Therefore, our paper could provide some biochemical evidences to comfirm that the new strain is a hybrid heterokaryon.
