摘 要:目的 :探讨兔应力性骨折 (SF)发生过程中 ,骨骼与肌肉形态学改变和肌匀浆磷脂酶A2 (PLA2 )活性的变化。方法 :14只雄性大白兔 ,随机分为对照组 4只 ,实验组 10只 ,置于模拟运动装置内使兔产生跑跳运动 ,每周训练 6d ,共 6周。结合骨闪烁显像诊断 ,于运动不同天数取样做骨、肌肉组织学检验。测定血清和肌肉过氧化脂质产物 (MDA)、PLA2 。结果 :(1)肉眼可见实验组病变部位骨骼较对照组突起、肿胀、触之较硬。光镜下病变部位骨骼肌细胞损害出现的时间较骨组织损害为早且随运动时间增加而增加。运动第 2~ 3周肌纤维损伤最为严重。 (2 )血清MDA较照组显著增加〔(0 .94± 0 .2 0 )mmol·L-1vs(0 .62± 0 .14)mmol·L-1,P <0 .0 5〕。肌肉组织MDA有增高趋势〔(1.0 6± 0 .2 7)mmol·gpro-1vs(0 .79± 0 .2 4 )mmol·gpro-1〕。 (3)腓肠肌匀浆磷脂酶A2显著高于对照组〔(3867± 5 83)mmol·s-1vs(2 65 0± 95 0 )mmol·s-1,P <0 .0 5 )〕。结论 :在应力性骨折发生过程中 ,骨骼肌组织细胞损害的时间早于骨组织 ,脂质过氧化物和磷脂酶A2Objective:To study the morphological changes of skeleton and muscle and phospholipase A 2 activity of muscular homogenate in the development of stress fracture in rabbits.Methods:14 male rabbits were divided into experimental group(n=10) and controlgroup(n=4).The experimental rabbits were stimulated to run and jump continually in a special device, 6 days per week for 6 weeks. Stress fracture was diagnosed by emission computcd axial tomograph(ECT). One or two rabbits were sacrificed every week for determining MDA and PLA 2 in skeletal muscle and serum, and for observing the pathological changes of muscle and bones under microscope.Results:(1)Gross inspection showed that the injured part was prominent, swollen and hardened. Under microscope, the lesion of skeletal muscle occurred earlier than that of bone tissue,and was aggravatcd as training time increased. The injury of the sural muscle was more severe in 2 to 3 weeks of the experiment. (2)Serum MDA of experimental rabbits were significantly higher than that of comtrol〔(0.94±0.20) mmol·L 1 vs (0.62±0.14)mmol·L 1 (P<0.05)〕,and the MDA in muscle were elevated too 〔(1.06±0.27)mmol·g pro 1 vs(0.79±0.24) mmol·g pro 1 〕.(3)The activity of phospholipase A 2 in sural muscle of experimental rabbits were increased significantly〔(3867±583)mmol·s 1 vs(2650±950)mmol·s 1 ,P<0.05〕.Conclusion:The changes of skeletal muscle injury were earlier than that of bone during the development of stress fracture, and the elevation of lipid peroxide as well as PLA 2 avtivity may promote the stress injury of skeletal muscle thus leading to stress fracture.
