摘  要:目的 :揭示局部脑缺血状态下热休克蛋白 (heatshockprotein,HSP)HSP70的表达与脑供血的关系,提供探讨针刺治疗脑梗塞研究的实验基础。方法:采用热凝法阻断大鼠一侧大脑中动脉(MCAo)造成局灶性脑缺血实验模型,采用DNA -RNA分子杂交技术研究了MCAo大鼠不同脑区HSP70的表达随缺血时间的改变。同时采用免疫组化的方法研究了HSP70蛋白表达的动态变化。结果:免疫阳性细胞出现较早并出现最多的区域是脑皮层区,纹状体次之,并多出现在缺血核心的周边区。分子杂交结果同样显示该模型中HSP70mRNA在大脑皮层中比在纹状体和海马中较早的出现且转录水平也高于纹状体和海马。结论:热休克蛋白HSP70因缺血被诱导,但其转录与表达又明显与供血状况有关。严重缺血区HSP70的表达受阻 ,提示改善脑局部微循环可促进HSP70的表达。Objective: To study the relationship between focal ischemia and expression of heat-shock proteins (HSP70) and to provide experimental data for study of acupuncture effect on ischemia. Methods:By using the method of thermocoagulation, to produce the middle cerebral arteria occlusion rat model. Using DNA-RNA hybridization, to study the changes of HSP70 gene transcription in different areas of rat brain and also determine the distribution of HSP70 protein in brain at different ischemia times. Results:Induced HSP70 primarily presents in cortex and striatum and mainly existed at the borders of infarction (in the penumbral areas). The molecular hybridization results show that HSP70 mRNA presented more and earlier in cortex than in striatum and hippocampus. Conclusion: HSP70 could be induced by ischemia. However, the expression of HSP70 is affected by the provision of blood. Long duration of middle cerebral artery ischemia that produces infarction does not induce HSP70.It suggested that by improving the blood microcirculation, HSP70 expression can be enhanced and improve the ability of thermotolerance of brain cells.
