摘  要:目的 研究全脑缺血再灌流后纹状体边缘区内一氧化氮合酶、神经肽Y等神经递质的表达变化。方法 在四管结扎造成的大鼠全脑缺血模型上,用组织化学和免疫组化方法观察纹状体边缘区内一氧化氮合酶、神经肽Y等阳性细胞的分布。结果 正常大鼠脑纹状体内有一氧化氮合酶及NPY阳性神经元的分布,全脑缺血再灌流后,边缘区中的NADPH-d阳性细胞明显增多。但缺血再灌流后第5天时阳性细胞数量回落,到第7天时恢复至正常对照组水平。全脑缺血再灌流7 d后,边缘区中NPY阳性反应消失,未见NPY阳性细胞。尾壳核中仍可见少量NPY阳性细胞。结论 纹状体边缘区中NOS阳性细胞对缺血有耐受性。全脑缺血后动物的学习记忆功能下降可能跟边缘区内NPY神经元缺失有关。 Objective To investigate the expression change of nitric oxide synthase and neuropeptide Y in the rat striatum following global ischemia. Methods Using histochemical and immunohistochemical methods, the distribution of nitric oxide synthase and neuropeptide Y in the rat striatum following global ischemia was studied in the 4-vessel ligation model. Results There are nitric oxide synthase and neuropeptide Y positive neurons in normal rat striatum. The number of NADPH-d positive neurons in the marginal division was increased obviously after global ischemia and reperfusion. However, the number of NADPH-d positive neurons in the marginal division was decreased 5 d after global ischemia and reperfusion, resuming the normal level on day 7. The NPY positive reaction in the marginal division disappeared 7 d after global ischemia and reperfusion. No NPY positive cells were found in the marginal division. A few NPY positive cells were observed in the caudate putamen. Conclusion The NOS positive neurons in the marginal division are more resistant to ischemic insult. The attenuation of learning and memory ability following global ischemia may involve the missing NPY positive neurons in the marginal division.
