巨噬细胞调理液对视网膜色素上皮细胞合成单核细胞趋化蛋白-1的作用

摘　　要：目的　评价巨噬细胞调理液 ( MCM)对视网膜色素上皮 ( RPE)细胞合成单核细胞趋化蛋白 - 1( MCP- 1)的作用 .方法　在培养的人 RPE细胞的培养液中加入 2 0 0 m L· L- 1培养人巨噬细胞调理液 ( MCM) ,培养 2 ,4 ,8,16,2 4和 4 8h后 ,对培养的细胞进行免疫组化和原位杂交 ,用 Western blot检测培养的上清液中 MCP- 1的含量 .同时在培养液中加入地塞米松 ( 10 - 8,10 - 7和 10 - 6 mol· L- 1 )和道诺霉素 ( 2 ,2 0和2 0 0 mg· L- 1 )重复上述实验 ,观察在此条件下 MCM对MCP- 1合成的作用 .结果　 2 0 0 m L· L- 1 MCM培养条件下RPE细胞 2 h时已检测出分泌的 MCP- 1并在 16h达到较高水平 .同时原位杂交和免疫组化在细胞中检测到 MCP- 1的表达 .加入地塞米松 ( P<0 .0 1)和道诺霉素 ( P<0 .0 5 )后 ,MCP- 1的分泌水平明显降低 ,其中地塞米松对 MCP- 1合成的抑制作用大于道诺霉素 .结论　 MCM刺激 RPE细胞产生MCP- 1,而 MCP- 1对巨噬细胞和淋巴细胞有较强的趋化作用 ,这种循环相互作用的过程对增生性玻璃体视网膜病变的发生可能有重要的作用 .在视网膜脱离后尽早使用地塞米松可能对抑制增殖性玻璃体视网膜病变有作用 .AIM To examine the effects of cultured macrophage conditioned media (MCM) on production and gene expression of monocyte chemotactic protein 1 (MCP 1) of human retinal pigment epithelial (HRPE) cell. METHODS HRPE were cultured with 200 mL·L -1 macrophage conditioned media (MCM) for 2, 4, 8, 24 or 48 h, and with additional dexamethasone (10 -8 , 10 -7 , 10 -6 mol·L -1 ) or daunorubicin (2, 20, 200 mg·L -1 ). Secreted MCP 1 in the supernatant of cultured RPE was measured using Western blot assay, and intracellular MCP 1 was detected by in situ hybridization and immunohistochemistry. RESULTS Secreted MCP 1 was detected with 200 mL·L -1 MCM for 2 hours after incubation, and reached the most intensity at 16 hours. MCP 1 in cytoplasm was noted during the same time. The levels of MCP 1 in cultures with Dexamethasone ( P <0.01) or Daunorubicin ( P <0.05) were decreased simultaneously. Dexamethasone showed a stronger effect than Daunorubicin. CONCLUSION MCM can stimulate RPE to produce MCP 1 which in turn can recruit macrophage into the inflammatory area. This may imply an loop interaction in the procedure of PVR. Early treatment with Dexamethasone may be helpful in preventing the occurrence of proliferative vitreoretinopathy.
